    Abstract: 本研究擬將91、92年度計畫中所開發出之聚丁基烷酯(PHB)多孔型薄膜之表面進行改質,予以固定幾丁聚醣,使薄膜適用於骨母細胞之培養,並探討其作為生醫材料之可行性。首先在薄膜表面進行化學修飾改質,將薄膜表面利用電漿聚合法接枝聚丙烯酸,然後分別以共價鍵結及塗佈吸附兩方法固定幾丁聚醣。共價鍵結法以1-ethyl-3-(3-diethylaminopropyl) carbodiimide hydrochloride (EDC)為活化劑,使幾丁聚醣之胺基與聚丙烯酸之羧基反應形成醯胺鍵,而固定在薄膜表面。塗佈吸收法乃將幾丁聚醣溶於丙烯酸水溶液中,均勻塗佈於薄膜上,靜置使丙烯酸擴散至薄膜中,經UV曝光聚合後使幾丁聚醣固定於薄膜表面,可藉由控制幾丁聚醣於丙烯酸中之濃度,使其厚度達到奈米層級。最後將幾丁聚醣/PHB複合薄膜用於骨母細胞培養,觀察對細胞之貼附與成長之影響,並以Escherichia coli.及Staphylococcus aureus進行之抗菌測試,期望能開發新型生物可分解性生醫膜材,以符合臨床之需求。
