    Abstract: 在許多的癌症都可發現上皮生長因子受體(EGFR)的基因放大(gene amplification)、過度表現(overexpression)或突變(gene mutation),包括大腸癌、肺癌、乳癌與頭頸癌等。我們已經證實了本土栽培的松杉靈芝(Ganoderma tsugae)萃取物對於多種的癌細胞株都有抑制生長的功效。此外,也利用投予松杉靈芝萃取物至接種A431細胞(人類上皮癌細胞株)的裸鼠實驗來偵測抑癌的效果。經免疫組織染色分析,初步的實驗結果顯示:松杉靈芝萃取物不僅會降低血管內皮細胞生長因子(VEGF)的表現量(附圖2),還會降低EGFR的蛋白質表現量(附圖3)。因此,本研究計畫的目標有下列四項: (1) 評估松杉靈芝萃取物是否可抑制內生性上皮生長因子受體蛋白的表現,進而抑制腫瘤細胞的生長、轉形與轉移等特性? (2) 探討松杉靈芝萃取物若可抑制上皮生長因子受體蛋白質表現之可能分子機轉。(3) 探討松杉靈芝萃取物是否藉由降低上皮生長因子受體的基因表現,因而造成癌細胞的凋亡與降低其血管新生的能力。 (4) 評估松杉靈芝萃取物是否可藉由抑制上皮生長因子受體的蛋白的表現,進而降低癌細胞對於化療的抗藥性。
