    摘要: 本實驗主要針對LL-AFS、LL-AYS與LL-AFVS等有機化合物與熱帶念珠菌(Candida tropicalis)進行抗菌活性測試,並利用原子力顯微術量測經過有機化合物LL-AFS、LL-AYS與LL-AFVS處理前後之表面形貌與生物力學結構變化。
    進行抑制熱帶念珠菌濃度測試發現,分別以LL-AFS、LL-AYS與LL-AFVS濃度300 ppm均可完全抑制熱帶念珠菌生長。故選擇濃度300 ppm之LL-AFS、LL-AYS與LL-AFVS與Candida念珠菌進行抑制時效測試。結果發現反應10分鐘即可有效抑制熱帶念珠菌生長。並探討有機化合物LL-AFS、LL-AYS與LL-AFVS處理前後之表面奈米結構與生物奈米力學變化,本篇研究選擇以原子力顯微術進行表面奈米結構與生物奈米力學分析。結果發現,熱帶念珠菌未經化合物表面結構處理之單一菌株長為2.67±0.54 μm、寬為1.08±0.32 μm、高度為0.30±0.04 μm;經化合物LL-AYS濃度300 ppm表面處理後,量測單一菌株長為2.07±0.67 μm、寬為0.94±0.23 μm、高度為0.53±0.14 μm。力學分析未經化合物處理前表面軟硬度為101.21±11.01 pN/nm;黏滯力為5.03±2.61 nN。經化合物LL-AYS(300 ppm)處理後,其表面軟硬度為117.24±10.53 pN/nm;黏滯力為17.66±1.89 nN。
    Candida tropicalis was treated with several organic compounds, and the differences of structural conformations and surface forces on Candida tropicalis were measured by atomic force microscope technology. Susceptibility to the minimal inhibitory concentration was investigated with the population analysis technique in the C. tropicalis strain.
    The experimental results showed that C. tropicalis were inhibited by organic compounds of LL-AFS, LL-AYS and LL-AFVS which their testing concentration were approximately equal or higher than 300 ppm of respective minimal inhibitory concentration(MIC).
    According to the susceptibility of LL-AFS, LL-AYS and LL-AFVS, there was a homogeneity of data with respective time inhibit curves showing a clear bactericidal effect after 10 minutes of exposure. Furthermore, a new drug will be designed by these results, susceptibility, respective time and force analysis.
