    Abstract: 醣類在細胞辨識、病毒感染及訊息傳導扮演十分重要的角色。醣類結構的定序,仍是具挑戰性研究主題之一。本論文藉由辨識、分析其基本的單糖結構組成,將其結果進ㄧ步的應用於醣類定序分析。在單糖上的羥基(-OH)之氫鍵和掌性結構之差異的前提下,藉SELEX(Systematic Evolution of Ligands by Exponential Enrichment)技術,專一性地篩選出特定DNA序列後,再進ㄧ步的研究DNA與單糖間的鍵結作用。本實驗以D-葡萄糖、D-甘露糖、D-半乳糖為研究目標,分析其與篩選出之DNA序列的鍵結。分別利用表面薄膜共振技術(Surface Plasmon Resonance)、凝膠移位分析法(Gel Mobility Shift Assay)、DNA熱變性曲線分析(Thermal Denaturation),研究其鍵結強弱。由凝膠移位分析法結果顯示適體會與單糖衍生物鍵結形成一複合物,且不受環境之pH值、離子強度及反應溫度所影響。由DNA熱變性曲線分析結果,證實有鍵結而造成曲線位移的現象。本研究結果說明DNA分子確實會與醣類作用。未來可望建立能辨識不同單糖的DNA序列資料庫,做為分析醣類序列的新方法,對DNA藥物及生物感測器的研發有相當大的發展潛力。
    Oligosaccharides sequence analysis is still a challenge topic of Glycobiology study. This study the binding of monosaccharides and aptamer which determined by SELEX (Systematic Evolution of Ligands by Exponential enrichment). With D-glucose, D-mannose, D-galactose as target molecular, To study the binding affinity between different monosaccharides and aptamer utilized SPR (Surface Plasmon Resonance) technology, GMSA (Gel Mobility Shift Assay), and Thermal Denaturation, respectively. From GMSA results, it’s found that the aptamer binding with the modified monosaccharides to form a complex. The interaction is not influenced by pH, ionic strength and temperature. It is also found that melting temperature curve shift in the result of Thermal Denaturation analysis. This result shows a evidence that binding of aptamer and monosaccharides. Can be expected to set up an DNA sequence database of distinguishing different monosaccharides in the future, make it for the new method of the Saccharides sequence analysis.
