    Abstract: 中草藥炮製是根據中醫臨床用藥理論和藥物配製的需要,對於藥材進行加工及處理,其主要的目的在於消除或降低藥材的毒性或副作用、改變藥性或提高療效、清除雜質和非藥用部分、便於煎服、製劑和貯藏等。
    本研究選擇了去皮半夏作為研究之對象,將其依據古籍記載之方法進行為期七天的炮製,利用固相萃取法(Solid Phase Extraction;簡稱SPE)進行樣品前處理,再藉由高效能液相層析儀(簡稱 HPLC)結合UV偵測器檢測上述之樣品,來建立中草藥半夏在炮製過程中化學成分變化的指紋圖譜。實驗中,我們利用RP-C18作為分離管柱,以兩段式gradient elution方式進行HPLC分離,偵測波長設定為210nm。實驗結果顯示,半夏中所含水溶性成分在炮製過程中均被浸泡至水中,其餘非水溶性成分則無明顯差異。由圖譜證實,SPE對於半夏藥材中所含之大多數成分均有濃縮效果。另外,本研究提供了半夏藥材炮製流程方法之規格化和科學化的參考依據。
    Processing of Chinese herbal medicine, is according to Chinese medicine theory and clinical need. Processing constituent of herbal medicine and multiplied in formulas containing more than one medicinal by the effects of processing. Its main purpose is eliminate or decrease the medicine toxicity or side effect, change drug’s nature or enhances the curative effect, elimination impurity and non-for medicinal part, easy to decocts medicine or stores it.
    In this study, we search the raw Pinellia ternate and processing by water containing 1﹪alum for 7 days, and its according to the traditional Chinese medical textbook. We used solid phase extraction (SPE) to prepare sample and detect by high permance liquid chromatography coupled ultraviolet/visible absorption detector (HPLC/
    UV). On the chemistry study of processed Pinellia ternate, we had developed the HPLC analysis methods and analyzed the fingerprint of Pinellia ternate before and after processing. In experiment, we used RP-C18 to be analyzing column, and used two program in gradient elution to separation, the detect wavelength is 210nm. Our results showed that, Pinellia ternate aqueous part was solute in water and non-aqueous part was no obviously difference. Identification by chromatography, SPE can condense most parts of Pinellia ternate. This study can provide establish the chemical fingerprints for raw Pinellia ternate, and establish the quality control criteria for each Pinellia ternate process products in order to produce consistent Pinellia ternate for appropriate medical use.
